The detection of Tramadol ( Generic Ultram ) and its metabolites in urine by chromatographic methods

The authors recommend detecting a compound drug Tramadol ( Generic Ultram ) and its metabolites in urinary extracts by thin-layer, gas-chromatographic, high-pressure liquid, and other chromatographic methods. The metabolites were identified with due consideration for their fragmentation during an electron strike thereof and their trimethylsilyl derivatives, mobility during separation in a thin layer, and results of selective extraction at various pH. The proposed scheme of biotransformation includes the formation of products of O- and N- demethylation, N, N- and N,O-didemethylation and hydroxylation of the cyclohexane fragment. The Rf metabolite values and their mass spectra are presented. During gas chromatographic analysis at an evaporator temperature at least 250 degrees C Tramadol ( Generic Ultram ) undergoes thermal destruction with the formation of water molecules and cyclohexane structure.

Tramadol ( Generic Ultram ) (zydol, Searle).

The account of Tramadol ( Generic Ultram ) (Zydol) in this month's issue is the first in a new series of articles reviewing new drugs and drugs which have recently become established in intensive and critical care nursing practice. It is intended through these articles to consider drugs from a nursing perspective and, in doing so, the key role of the nurse in evaluating the appropriateness, efficacy and safety of drug therapy is readily acknowledged. The format provides a brief description of dosage and available dosage forms together with pharmaceutical aspects including preparation, administration and storage. The actions and clinical uses of drugs are also discussed and summarised in a brief therapeutic comment. The Editor is anxious that 'Drug Therapy Review' serves a useful and practical function within the nursing process and the comments of our readers, including suggestions on topics for future review, will be gratefully received.

Opioid neurotoxicity: comparison of morphine and Tramadol ( Generic Ultram ) in an experimental rat model.Atici S, Cinel L, Cinel I, Doruk N, Aktekin M, Akca A, Camdeviren H, Oral U.Department of Anesthesiology & Reanimation, Mersin University School of Medicine, Mersin, Turkey. Histopathologic changes in rat brain due to chronic use of morphine and/or Tramadol ( Generic Ultram ) in progressively increased doses were investigated in this study. Thirty male Wistar rats (180-220 g) were included and divided into three groups. Normal saline (1 ml/kg) was given intraperitoneally as placebo in the control group (n = 10). Morphine group (n = 10) received morphine intraperitoneally at a dose of 4 mg/kg/day for the first 10 days, 8 mg/kg/day between 11-20 days, and 12 mg/kg/day between 21-30 days. The Tramadol ( Generic Ultram ) group (n = 10) received the drug intraperitoneally at doses of 20, 40, and 80 mg/kg/day in the first, second, and the third 10 days of the study, respectively. All rats were decapitated on the 30th day and the brain was removed intact for histology. The presence and the number of red neurons, which are a histologic marker of apoptosis, were investigated in the parietal, frontal, temporal, occipital, entorhinal, pyriform, and hippocampal CA1, CA2, CA3 regions. Red neurons were found in morphine and Tramadol ( Generic Ultram ) groups but not in the control group. The total number of red neurons was not different in morphine and Tramadol ( Generic Ultram ) groups, but the numbers of red neurons were significantly higher in the temporal and occipital regions in Tramadol ( Generic Ultram ) group as compared with the morphine group (p < .05). In conclusion, chronic use of morphine and/or Tramadol ( Generic Ultram ) in increasing doses is found to cause red neuron degeneration in the rat brain, which probably contributes to cerebral dysfunction. These findings should be taken into consideration when chrome use of opioids is indicated.

Intrathecal injection of codeine, buprenorphine, tilidine, Tramadol ( Generic Ultram ) and nefopam depresses the tail-flick response in rats.

The effect of intrathecal (i.t.) injection of the analgesic agents, codeine, buprenorphine, tilidine and one of its metabolites, nortilidine, Tramadol ( Generic Ultram ) and nefopam, was determined in the tail-flick test performed on rats. ED50 values were derived from the dose-response lines. The relative potency ranking established from the ED50 values is buprenorphine (0.4 nM) greater than nortilidine (29 nM) = Tramadol ( Generic Ultram ) (26 nM) = nefopam (34 nM) greater than codeine (42 nM) greater than tilidine (118 nM). An i.t. injection of the opiate antagonist, naloxone (5 micrograms), prevented the antinociceptive effect of all analgesic agents administered at the highest dose tested. It is concluded that these analgesic agents, like morphine, exert their effect at least in part through a spinal site of action.

Effects of Tramadol ( Generic Ultram ) on haemodynamics and blood gases in the early postoperative period

A variety of opioids is available for treatment of acute pain. Sometimes administration is limited due to typical side effects such as respiratory depression or pressure increase in the pulmonary circulation. Tramadol ( Generic Ultram ), a synthetic opioid, was investigated in a dosage of 1.5 mg/kg body weight i.v. with regard to changes in haemodynamic parameters and in blood gases. The haemodynamic parameters generally remained stable; all changes were statistically non significant. There were no signs of respiratory depression. The risk of pain therapy with opioids seems to be reduced further by the introduction of this agent.

Use of Tramadol ( Generic Ultram ) versus pethidine versus denaverine suppositories in labor--a contribution to noninvasive therapy of labor pain

Because Tramadol ( Generic Ultram ) does not exhibit an depressive effect on ventilatory activity it is often be used in the obstetrical analgesia, at most in form of an intramuscularly injection. In a prospective study on at all 49 women under labour the clinical effect of the noninvasive rectal application of Tramadol ( Generic Ultram ), Pethidin, and Denaverin has been compared. The first dosage was 100 mg of all substances. Around the half of the women said that analgetic effect was good or very good. On only every fifth it was sufficient or not enough. The effect was at near the same in all treatment groups. Because of a low incidence of maternal side effects, the absence of side effects on the newborn, and near the same results on the analgetic effect of parenteral application in other studies, Tramadol ( Generic Ultram ) suppositories can be recommended for obstetrical analgesia.
